PFDN6 is a β-subunit of the heterohexameric prefoldin chaperone complex that captures non-native actin and tubulin via its tentacle tips and delivers them to the cytosolic chaperonin TRiC/CCT for productive folding (PMID:9630229, PMID:14634002). When not incorporated into the hexamer, monomeric PFDN6 is degraded by the ubiquitin–proteasome system, and co-assembly with partner subunits provides mutual stabilization (PMID:21478150). PFDN6 also serves as a β-subunit of the URI prefoldin-like complex and selectively associates with the URI/Bud27 α-subunit (PMID:30484155, PMID:30484156). Loss of PFDN6 in cancer cells attenuates proliferation and migration through impaired AKT signalling and altered transcriptional regulation of ZNF575, while in Drosophila neurons its depletion exacerbates Tau-mediated neurodegeneration (PMID:36300673, PMID:39944471, PMID:41533771).
