Synthesis pass · prose summary of the discoveries below

PES1 (Pescadillo) is a nucleolar protein essential for large ribosomal subunit biogenesis and with expanding roles in telomere maintenance, estrogen receptor signaling, and immune evasion. It assembles with Bop1 and WDR12 into the PeBoW complex, which is required for processing of 32S/36S precursor rRNA into mature 28S and 5.8S rRNA; its BRCT domain is indispensable for nucleolar localization, PeBoW incorporation, and rRNA processing function, and disruption of the complex triggers p53 accumulation and cell cycle arrest (PMID:15225545, PMID:16043514, PMID:17189298). Beyond ribosome biogenesis, PES1 directly interacts with TERT to promote telomerase RNA–TERT assembly and telomerase activity (PMID:31106266), differentially stabilizes ERα while destabilizing ERβ through CHIP-mediated ubiquitination—a process counteracted by SUMO-1/2/3 modification at K517 (PMID:22820289, PMID:27409667)—and suppresses CD8⁺ T cell–mediated immune surveillance by disrupting the ILF3–IL15 mRNA stabilization complex (PMID:36959575). PES1 protein stability is itself regulated by Trim23-mediated ubiquitination, which is inhibited by the lncRNA SNHG17 (PMID:34782005).

2000 The murine Pes1 protein contains a BRCT (BRCA1 C-terminal) domain and localizes to the nucleus when expressed recombinantly in HepG2 cells, establishing it as a nuclear protein with a domain typically found in cell-cycle checkpoint and DNA repair factors. Recombinant protein expression, nuclear localization by microscopy, sequence analysis Genomics Medium 11112348
2004 Pes1 physically interacts with the nucleolar protein Bop1; this interaction is essential for the efficient incorporation of Pes1 into nucleolar preribosomal complexes, and both proteins direct common pre-rRNA processing steps (28S and 5.8S rRNA maturation). Pes1 mutants defective in Bop1 binding lose the ability to affect rRNA maturation and arrest the cell cycle. Co-immunoprecipitation, dominant-negative transposon-derived mutants, pre-rRNA processing analysis, cell cycle assays Molecular cell High 15225545
2005 Pes1 and Bop1 form a stable trimeric complex with the novel WD40 repeat protein WDR12 (the PeBoW complex). Endogenous WDR12 is required for processing of 32S precursor rRNA and cell proliferation; a dominant-negative WDR12 mutant blocks rRNA processing and induces reversible cell cycle arrest, accumulating p53 in a p19ARF-independent manner in proliferating but not quiescent cells. Co-immunoprecipitation, conditional dominant-negative expression, rRNA processing assays, p53 accumulation assays The Journal of cell biology High 16043514
2005 In yeast, the Pes1 orthologs Ytm1, Nop7, and Erb1 form a heterotrimeric subcomplex within 66S preribosomes. Ytm1 binds directly to Erb1; mutations in Ytm1 WD40 motifs disrupt Erb1 binding, destabilize the heterotrimer, and delay pre-rRNA processing and nuclear export of preribosomes. Genetic analysis, co-immunoprecipitation, sucrose gradient fractionation, mutant analysis of WD40 motifs Molecular and cellular biology High 16287855
2006 Dominant-negative Pes1 mutants with N- or C-terminal truncations (M1 and M5) co-immunoprecipitate Bop1 and WDR12, incorporate into large preribosomal complexes, block 36S/32S to 28S rRNA processing, inhibit cell proliferation, and induce p53 accumulation in proliferating but not resting cells, confirming that the dominant-negative effect is mediated through the intact PeBoW complex. Co-immunoprecipitation, sucrose gradient sedimentation, rRNA processing analysis, flow cytometry, p53 Western blot Nucleic acids research High 16738141
2006 The BRCT domain of Pes1 is essential for nucleolar localization and rRNA processing function. Deletion of the BRCT domain or point mutations of conserved residues cause diffuse nucleoplasmic distribution of Pes1, failure to replace endogenous Pes1 in rRNA processing, reduced protein stability, and failure to incorporate into the PeBoW complex. Conditional siRNA knock-down/knock-in system, panel of truncation mutants, immunofluorescence, rRNA processing assays Nucleic acids research High 17189298
2007 Assembly and stability of the PeBoW complex require Bop1 as the integral central component: recombinant co-expression of all three subunits (Pes1, Bop1, WDR12) is sufficient for complex formation; Bop1 knockdown abolishes co-purification of Pes1 with WDR12; Bop1 transport from cytoplasm to nucleolus is Pes1-dependent, while Pes1 can migrate to the nucleolus and bind preribosomal particles independently of Bop1. Recombinant co-expression, co-immunoprecipitation, siRNA knockdown, indirect immunofluorescence, cell fractionation, sucrose gradient centrifugation Molecular and cellular biology High 17353269
2007 Light chain 1 of microtubule-associated protein 1B (Mtap1b-LC1) interacts with Pes1 specifically through the Pes1 BRCT domain (identified by yeast two-hybrid and confirmed by co-immunoprecipitation). Overexpression of Mtap1b-LC1 redistributes Pes1 from the nucleus/nucleolus to the cytoplasm and reduces nuclear Pes1, resulting in diminished cell proliferation, mimicking the effect of Pes1 depletion by shRNA. Yeast two-hybrid, co-immunoprecipitation, deletion analysis, protein localization studies, cell proliferation assays, shRNA The Journal of biological chemistry High 17308336
2008 In yeast, Ytm1, Erb1, and Nop7 (orthologs of WDR12, Bop1, Pes1) assemble into preribosomes in an interdependent manner. Specific domains within each protein mediate pairwise interactions and are sufficient for recruitment into preribosomes; dominant-negative truncations of each reveal distinct interaction and functional domains. Genetic epistasis, truncation mutagenesis, co-immunoprecipitation, dominant-negative growth assays Molecular biology of the cell High 18448671
2009 The nucleolar marker protein B23 (nucleophosmin) physically interacts with PES1 in vivo (confirmed by co-immunoprecipitation of endogenous proteins) and in vitro (pulldown assay with mapped interaction domains). Knockdown of B23 by RNAi increases the nucleoplasmic distribution of PES1, indicating that B23 is involved in nucleolar localization of PES1. 2012 PES1 differentially regulates ERα and ERβ protein stability in breast cancer: PES1 enhances ERα transcriptional activity and increases ERα protein stability while inhibiting ERβ transcriptional activity and decreasing ERβ protein stability through the ubiquitin-proteasome pathway mediated by the co-chaperone CHIP. PES1 also transforms normal human mammary epithelial cells and is required for estrogen-induced breast tumor growth in nude mice. Co-immunoprecipitation, ubiquitination assays, luciferase reporter assays, siRNA knockdown, soft-agar transformation assay, xenograft mouse model The Journal of clinical investigation High 22820289
2012 c-Jun transcriptionally activates PES1 expression in colon cancer by directly binding to the -274/-264 region of the PES1 promoter; JNK signaling is upstream of c-Jun in controlling PES1 levels. This was validated by chromatin immunoprecipitation and EMSA. Luciferase reporter assays, chromatin immunoprecipitation (ChIP), electrophoretic mobility shift assay (EMSA), promoter deletion analysis, siRNA knockdown PloS one High 22860098

2016 PES1 is SUMOylated at lysine K517 in its C-terminal Glu-rich domain by SUMO-1, -2, and -3, with SUMOylation stimulated by estrogen. SUMOylation stabilizes PES1 by inhibiting its ubiquitination, enhances PES1-mediated promotion of ERα transcriptional activity and ERα stability, and increases the proportion of S-phase cells, promoting breast cancer cell proliferation in vitro and in vivo. SUMO modification assays, site-directed mutagenesis (K517R), ubiquitination assays, luciferase reporter assays, cell cycle analysis, xenograft mouse model Oncotarget High 27409667
2018 PES1 is transcriptionally regulated by BRD4 in hepatocellular carcinoma and promotes glycolysis by altering expression of metabolic genes (GLUT1, PKM2, ENO1, FBP1, PCK1).
