IK (also called RER) encodes a 19 kDa cytokine secreted by leukemic and cancer cell lines that inhibits IFN-gamma-induced HLA class II MHC antigen expression, identified through protein purification, cDNA cloning, and antibody-mediated neutralization of its biological activity (PMID:7970704). Mechanistically, IK suppresses class II MHC expression by decreasing levels of the class II MHC transcription activator (CIITA), and a truncated form translated from an internal methionine (tIK) retains both secretion and this inhibitory activity (PMID:17075801). In MRL/lpr mice, tIK treatment reduces renal damage, T cell and macrophage infiltration, IFN-gamma and IL-2 production, and anti-DNA antibody titers, establishing that IK modulates autoimmune pathology through class II MHC-dependent suppression of immune activation (PMID:17075801). Beyond its role as an immunomodulatory cytokine, no further molecular mechanism of CIITA suppression or receptor engagement has been characterized in the available corpus.
