Tamalin (GRASP) is a neuronal PDZ domain-containing scaffold protein that assembles multi-protein complexes at the plasma membrane to control receptor trafficking and GTPase signaling (PMID:11850456, PMID:10828067). Through its PDZ domain it directly binds the class I PDZ-binding motifs of group 1 metabotropic glutamate receptors (mGluR1a, mGluR5), while its C-terminal half engages cytohesin-family ARF-GEFs; native mGluR1a/tamalin/cytohesin-2 ternary complexes exist in brain, and tamalin promotes intracellular trafficking and cell-surface expression of group 1 mGluRs (PMID:11850456). Crystallographic analysis shows tamalin self-assembles into autoinhibited oligomers via reciprocal PDZ/intrinsic-ligand interactions, an assembly competitively disrupted by mGluR binding that liberates the intrinsic ligand to facilitate receptor surface trafficking (PMID:17396155). Beyond mGluRs, tamalin nucleates a broader postsynaptic scaffold network, binding SAPAP/GKAP, S-SCAM, PSD-95, and Mint2/X11β through distinct domains (PMID:12586822). It couples receptor signaling to small-GTPase cascades: it bridges the ARF-GEF ARNO/cytohesin-2 to the Rac-GEF Dock180, driving ARF-to-Rac crosstalk that promotes cell migration (PMID:20016009, PMID:23441967), and a ligand-dependent TrkCT1–tamalin complex activates Rac1 via ARF6 to produce membrane ruffling and protrusions (PMID:16636148). Tamalin also carries an ITAM whose paired tyrosines are phosphorylated by c-Src and Fyn to recruit and activate Syk kinase (PMID:15173175). In vivo, tamalin is required for acute and adaptive behavioral responses to opioids and psychostimulants (PMID:17766434) and for electroconvulsive-shock–induced hippocampal neurogenesis, mossy fiber sprouting, and dendritic arborization (PMID:22396401).
