Emerin (EMD) is an inner nuclear membrane LEM-domain protein that anchors to the nuclear envelope via a C-terminal hydrophobic domain and functionally cooperates with A-type lamins to maintain nuclear integrity and regulate signaling in striated muscle (PMID:7894480, PMID:17536044). Loss-of-function mutations in EMD cause X-linked Emery–Dreifuss muscular dystrophy (EDMD), and emerin deficiency activates the ERK1/2 MAPK cascade in cardiomyocytes, a pathway shared with LMNA-mutant EDMD and implicated in cardiomyopathy pathogenesis (PMID:17567779). Beyond its structural role, emerin is phosphorylated on its LEM domain by PRKACA downstream of C16-ceramide signaling, enabling it to bind LC3B and promote autophagosome formation (PMID:24819607). Emerin is also phosphorylated by virion-associated ERK2 during lentiviral infection, facilitating nuclear import and chromatin integration of the pre-integration complex (PMID:20463147).
