EMD

Emerin (EMD) is an inner nuclear membrane LEM-domain protein that anchors to the nuclear envelope via a C-terminal hydrophobic domain and functionally cooperates with A-type lamins to maintain nuclear integrity and regulate signaling in striated muscle (PMID:7894480, PMID:17536044). Loss-of-function mutations in EMD cause X-linked Emery–Dreifuss muscular dystrophy (EDMD), and emerin deficiency activates the ERK1/2 MAPK cascade in cardiomyocytes, a pathway shared with LMNA-mutant EDMD and implicated in cardiomyopathy pathogenesis (PMID:17567779). Beyond its structural role, emerin is phosphorylated on its LEM domain by PRKACA downstream of C16-ceramide signaling, enabling it to bind LC3B and promote autophagosome formation (PMID:24819607). Emerin is also phosphorylated by virion-associated ERK2 during lentiviral infection, facilitating nuclear import and chromatin integration of the pre-integration complex (PMID:20463147).

   Two studies addressed the functional relationship between emerin and lamin A/C: digenic analysis in a human family demonstrated synergistic interaction between EMD and LMNA mutations for full EDMD expression, while expression profiling of Emd-null mouse hearts revealed aberrant activation of the ERK1/2 MAPK pathway — identifying a specific signaling defect downstream of emerin loss that is shared with Lmna-mutant cardiomyopathy.

   Two studies in 2014 expanded emerin's functional and phenotypic scope: phosphorylation of emerin's LEM domain by PRKACA in response to C16-ceramide was shown to enable LC3B binding and enhanced autophagosome formation, while identification of an EMD deletion causing severe dilated cardiomyopathy with minimal skeletal involvement demonstrated that emerin loss-of-function extends beyond classic EDMD to isolated DCM.
