COMMD6 is a COMM domain-containing protein that serves as an obligate structural component of the CCC (COMMD/CCDC22/CCDC93) complex, essential for the stability of all ten COMMD family members and the complex core subunits CCDC22, CCDC93, and C16orf62 (PMID:29545368). Loss of COMMD6 in hepatocytes destabilizes the CCC complex, impairing endosomal recycling of LDLR and ATP7B, which leads to elevated plasma LDL cholesterol, exacerbated atherosclerosis, and hepatic copper accumulation under copper challenge (PMID:29545368, PMID:33262129). COMMD6 also inhibits TNF-induced NF-κB activation through its COMM domain, where residues Trp24 and Pro41 are critical for this inhibitory function; unlike COMMD1, COMMD6 does not bind IκBα, indicating a partially distinct mechanism of NF-κB suppression mediated through direct interaction with RelA (PMID:16573520).
