REX1 binds the unmethylated allele of imprinting control regions (Peg3, Gnas) to protect them from DNA methylation, and represses Xist transcription at its regulatory region; REX1 is the critical substrate of the X-encoded E3 ubiquitin ligase RNF12, whose ubiquitination targets REX1 for proteasomal degradation to permit Xist upregulation and X-chromosome inactivation (PMID:21233130, PMID:22596162, PMID:30420655). REX1 also drives mitochondrial fission and glycolytic metabolism by activating cyclin B1/B2 transcription, leading to CDK1-dependent DRP1 phosphorylation (PMID:23939908).
