At tight junctions, YBX3 is sequestered by ZO-1/ZO-2 and regulated by competing interactions with CDK4, RalA, GEF-H1/RhoA, Apg-2, and symplekin; release from this junctional pool permits nuclear translocation, where YBX3 activates proliferative gene promoters (cyclin D1, PCNA) and represses differentiation-associated targets (megalin, cubilin, AML1, claudin-5, occludin) (PMID:12566432, PMID:16508013, PMID:19328795, PMID:20133480, PMID:26106824). As an RNA-binding protein, YBX3 directly binds and stabilizes mRNAs encoding amino acid transporters (SLC7A5, SLC3A2, SLC1A5) and metabolic regulators (Pparg), thereby maintaining intracellular amino acid pools and supporting processes including brown adipose thermogenesis and skeletal muscle differentiation (PMID:31189097, PMID:38159852, PMID:39481849). In vivo, tissue-specific loss of Ybx3 impairs brown fat thermogenesis and exacerbates diet-induced obesity, while in kidney it reduces fibrotic remodeling, underscoring context-dependent roles in metabolic homeostasis and tissue injury (PMID:39481849, PMID:37408260).
