Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

SNRPG (SmG) is an Sm-domain spliceosome-associated protein that participates in alternative splicing regulation and has been repeatedly implicated as a driver of cancer cell proliferation and metastasis (PMID:32296580, PMID:41526677). In germ cells, SNRPG physically associates with PCID2, hnRNPH1, and SF3B1 within a PCID2-containing splicing complex that modulates alternative splicing during spermatogonial differentiation (PMID:41526677). Beyond its splicing-complex role, SNRPG operates in oncogenic signaling through two distinct routes: in glioblastoma it acts upstream of Myc in a p53-dependent pathway, where its loss arrests the cell cycle, lowers Myc protein, and sensitizes temozolomide-resistant cells to chemotherapy (PMID:32296580); in NSCLC it directly binds the transcription factor WT1 and destabilizes it, preventing WT1 from activating CDK14 transcription and thereby restraining epithelial-mesenchymal transition and metastasis downstream of 5-HT signaling (PMID:41016985). SNRPG also engages in direct, high-affinity physical interaction with the RING finger domain of RBBP6 (KD ~3 nM) (PMID:34956492). A unifying biochemical mechanism connecting its splicing function to its transcription-factor-destabilizing activity has not been characterized in the available corpus.

2020 SNRPG knockdown via shRNA in glioblastoma cells caused cell cycle arrest and reduced Myc protein levels; the effect of SNRPG loss on p53 levels and cell cycle progression was Myc-dependent, placing SNRPG upstream of Myc in a p53-dependent signaling pathway. SNRPG was also elevated in TMZ-resistant GBM cells, and its downregulation sensitized resistant cells to temozolomide via the p53 signaling pathway. shRNA knockdown, gene expression profiling, Western blot, xenograft tumor model Cancer biology & medicine Medium 32296580

2025 5-HT inhibits SNRPG expression in NSCLC cells; SNRPG in turn directly binds to WT1 protein and negatively regulates its protein levels, preventing WT1 from binding the CDK14 promoter. Loss of SNRPG leads to WT1 accumulation, CDK14 transcriptional activation, and promotion of EMT and metastasis. This defines a 5-HT/SNRPG/WT1/CDK14 signaling axis. 1997 Antisense RNA-mediated suppression of CaSm (an Sm-like protein with ~60% similarity to Sm-G/SNRPG) reduced the ability of pancreatic cancer cells to form colonies in soft agar, establishing that CaSm expression is required for maintenance of the transformed state. Note: CaSm is a distinct Sm-like protein, not SNRPG itself, but this paper identifies SNRPG as the closest human paralog for context.
