RIIα physically binds AKAP1 to enforce cytoplasmic anchoring of PKA, and its translocation with PKA-C from cytoplasm to nucleus precedes meiotic resumption in oocytes, establishing RIIα as a determinant of PKA compartmentalization (PMID:24501172, PMID:22674394, PMID:23426434); an analogous role in neurons directs dendritic localization of PKA catalytic subunits in medial habenula neurons, where loss of Prkar2a reduces local PKA activity and alters feeding and exercise behaviors that are rescued by habenular re-expression (PMID:33141766). Because RIIα restrains catalytic activity, its loss produces unrestrained cAMP/PKA signaling: Prkar2a-deficient mice develop histiocytic sarcomas, and reduction of PKA type II activity by Prkar2a haploinsufficiency rescues the dysplastic bone phenotype of Prkar1a heterozygotes (PMID:26608815, PMID:26246497). In liver, a high PRKACA/PRKAR2A ratio activates PKA and confers fibrolamellar-like features, and combined BAP1 and PRKAR2A loss drives hepatocyte transdifferentiation into ductal/progenitor-like cells, with PRKAR2A loss specifically required to overcome cell-cycle arrest (PMID:31862487, PMID:37137901). Beyond PKA regulation, RIIα directly binds the IFN-γ receptor transmembrane domain downstream of DP1 activation to suppress JAK2/STAT1-driven M1 macrophage polarization and promote inflammatory resolution (PMID:27621415), and the PRKAR2A promoter is transcriptionally repressed by HDAC1/2–Sp1/Sp3–RbAp48 complexes at GC-rich domains (PMID:19818097).
