Its biology centers on engagement of cell-surface receptors and activation of downstream signaling: WFDC2 binds the extracellular domain of EGFR and, depending on context, modulates EGFR/AKT/GSK3B/Snail signaling and epithelial-mesenchymal transition (PMID:32678075), while in ovarian cancer it drives EMT and invasion through PI3K/AKT-dependent induction of MMP-2 (PMID:31118763) and activation of JAK/STAT3 signaling (PMID:31477564). WFDC2 also forms a cytoplasmic and membrane-associated ternary complex with ANXA2 and MMP-2, using ANXA2 as a bridge to promote migration (PMID:31210752).
