On the male-promoting side it directly activates Sox9b in tilapia (PMID:30415011), cooperates with Sf1 to induce gsdf (PMID:27027772), and behaves as a pioneer factor that opens chromatin and recruits SOX9 to newly accessible regions to consolidate Sertoli-cell fate (PMID:34096593). On the repressive side it silences the female determinant Foxl2 — sufficient, even in adult ovary, to reprogram granulosa cells into Sertoli-like cells — and in germ cells directly represses the pluripotency factors Sox2, Esrrb, and Nr5a2/Lrh1 to prevent teratoma formation (PMID:21775990, PMID:25683803, PMID:20007774, PMID:23473982). Its own expression is driven in Sertoli cells by GATA4 and by Sp1/Sp3/Egr1 elements, modulated by FSH/cAMP-PKA signaling, and its DM-domain NLS mediates importin-β1-dependent nuclear import (PMID:14673170, PMID:11870074, PMID:11181532, PMID:17459496).
