BRAP (also IMP, BRAP2, RNF52) is a cytoplasmic RING-domain E3 ubiquitin ligase that sets activation thresholds for mitogenic and stress-signaling cascades (PMID:14724641). Its founding function is as a Ras-responsive brake on the Raf-MEK-ERK pathway: BRAP inhibits the scaffold KSR1 by disrupting KSR1 homo-oligomerization and B-Raf/c-Raf hetero-oligomerization, thereby limiting MEK recruitment and c-Raf activation and acting as a threshold modulator rather than altering response timing (PMID:14724641, PMID:18332145). Ras activation drives BRAP auto-polyubiquitination and autodegradation, relieving this inhibition and permitting productive Raf-MEK assembly (PMID:14724641, PMID:19091743); BRAP stability is in turn opposed by the deubiquitylases USP15 and USP4, which counter its autoubiquitination (PMID:23105109). In the developing CNS, BRAP partners with the Nde1-Lis1 complex to tune MAPK thresholds during neural progenitor proliferation and differentiation (PMID:23673330), and BRAP-mediated polyubiquitination of Skp2 restrains G1/S transition, with its loss causing premature S-phase entry and microcephaly (PMID:28768199). Independently of catalysis-driven signaling, BRAP serves as a cytoplasmic retention factor that binds nuclear-localization signals in a phosphorylation-dependent manner to sequester nuclear proteins including p21/CDKN1A, HMG20A, NuMA1, and certain viral cargoes such as SV40 large T antigen (PMID:15340083, PMID:20040518, PMID:23707952). BRAP also restrains NF-κB nuclear translocation through interaction with IKK signalosome components IκBβ and IKKβ and with Nedd8/SCF complexes (PMID:21670849, PMID:23554956), and its loss in mouse brain produces persistent DNA double-strand breaks and elevated histone H2A ubiquitination linked to senescence and neurodegeneration (PMID:35754718). BRAP variants are genetically associated with myocardial infarction (PMID:19198608).

2004 BRAP/IMP (Impedes Mitogenic signal Propagation) is a Ras-responsive E3 ubiquitin ligase that inhibits KSR (a scaffold/adaptor protein), thereby limiting functional assembly of Raf-MEK complexes and suppressing MAP kinase cascade activation. Upon Ras activation, IMP undergoes auto-polyubiquitination, which releases its inhibition of Raf-MEK complex formation. IMP depletion increases stimulus-dependent MEK activation without altering timing or duration of the response, establishing IMP as a threshold modulator. siRNA depletion, ubiquitination assays, epistasis with Ras, Co-IP, MEK activation assays Nature High 14724641
2004 BRAP2 directly interacts with the cell cycle inhibitor p21 in vitro and in vivo through the nuclear localization signal (NLS) of p21 and the C-terminal portion of BRAP2. Co-transfection of BRAP2 causes p21 to localize to the cytoplasm. During monocytic differentiation of U937 and HL-60 cells, BRAP2 expression is upregulated concomitantly with cytoplasmic relocalization of p21, indicating BRAP2 acts as a cytoplasmic retention protein for p21. Co-immunoprecipitation in vitro and in vivo, co-transfection localization assays, immunoblotting during monocyte differentiation Molecular and cellular biology Medium 15340083
2008 IMP inhibits signal propagation through Raf to MEK by disrupting KSR1 homo-oligomerization and B-Raf/c-Raf hetero-oligomerization, thereby impairing both MEK recruitment to activated Raf and c-Raf kinase activation. Human KSR1 promotes assembly of multivalent Raf-MEK complexes required for c-Raf kinase activation, and IMP exploits this property to modulate signal amplitude. Co-immunoprecipitation, KSR1 oligomerization assays, kinase activity assays, IMP expression/depletion The Journal of biological chemistry Medium 18332145
2008 IMP concentrations are directly regulated by Ras through induction of IMP autoubiquitination (autodegradation), permitting productive Raf-MEK complex assembly. The mechanism of inhibition involves KSR inactivation as the key downstream target. 2011 BRAP knockdown in human aortic smooth muscle cells attenuated cell proliferation and reduced secretion of MCP-1 and IL-8 in response to LPS, and prohibited NF-κB nuclear translocation without affecting NF-κB protein levels. Co-immunoprecipitation confirmed interaction between BRAP and IκBβ and IKKβ, two major components of the IKK signalosome. siRNA knockdown, co-immunoprecipitation, NF-κB nuclear translocation assay, cytokine ELISA Molecular medicine (Cambridge, Mass.) 2012 BRAP/IMP E3 ubiquitin ligase interacts with deubiquitylases USP15 and USP4 through the N-terminal DUSP-UBL domain of USP15 and the coiled-coil region of BRAP. USP15 and USP4 oppose BRAP autoubiquitination, while BRAP promotes USP15 ubiquitylation. USP15 depletion destabilizes BRAP via proteasomal degradation; catalytically active USP15 rescues BRAP levels. USP15 depletion decreases MAPK signaling amplitude in response to EGF and PDGF, in part through decreased CRAF levels. 2013 Brap directly interacts with Lis1, and a Nde1-Lis1-Brap complex regulates MAPK signaling thresholds in the developing mammalian CNS. Nde1-Lis1 deficiency causes spatially dependent alterations of the MAPK scaffold KSR and hyperactivation of MAPK. Epistasis analyses supported synergistic Brap and Lis1 functions in controlling neural progenitor proliferation and differentiation. 2013 Brap2 is a novel Nedd8-binding protein that interacts with the SCF ubiquitin ligase complex and is involved in NF-κB nuclear translocation following TNF-α stimulation. A putative neddylation site in Brap2 is associated with NF-κB activity. Brap2 modulates temporal control of NF-κB nuclear translocation mediated by inflammatory response. 2009 BRAP2 inhibits nuclear import of specific viral proteins (SV40 large T antigen and HCMV ppUL44) in a manner dependent on phosphorylation sites flanking their nuclear localization signals. Substitution of CDK phosphorylation site T124 of T-ag with Ala prevented BRAP2 inhibition, while Asp (phosphomimetic) enhanced it. Direct high-affinity binding of BRAP2(442-592) to T-ag(111-135) was strictly dependent on negative charge at T124 and the NLS, demonstrated by pulldown/AlphaScreen assays. No effect of BRAP2 was observed on HSV-1 pUL30 (which lacks a phosphorylation site near its NLS) or HIV-1 Tat. 2017 Loss of Brap in mouse neural progenitor cells results in premature G1/S phase transition due to elevated Skp2 (SCF ubiquitin ligase component), which increases p27Kip1 destruction. Brap-mediated polyubiquitination of Skp2 was identified as a key regulatory step. Loss of Brap led to elevated Skp2, shortened G1 phase, premature S phase entry, hindered S phase progression, increased G2 arrest, impeded neuronal differentiation, and microcephaly. 2018 In C. elegans, BRAP-2 promotes DNA damage-induced germline apoptosis by attenuating pro-survival signals of AKT-1 and PMK-1/SKN-1. Loss of brap-2 reduces germline apoptosis after DNA damage and increases skn-1 mRNA. The interaction between BRAP-2 and the AKT-1 phosphatase (C. elegans homolog of PHLPP1/2) is conserved. SKN-1 RNAi or loss of pmk-1 in brap-2 mutants restored apoptosis to wild-type levels.
