Beyond its ligase role, TRIM27 serves as an endosomal scaffold at retromer-positive structures—recruiting gp130/JAK1/STAT3 for IL-6 signaling and regulating WASH-dependent actin dynamics under the control of CLEC16A and WNK1—and is itself stabilized by the deubiquitinase USP7 within the non-canonical PRC1.1 Polycomb complex, where USP7 counteracts TRIM27 autoubiquitination to maintain chromatin occupancy and H2AK119ub levels (PMID:30143645, PMID:37307465, PMID:36538041, PMID:34113809).
