Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

SNTB1 (beta-1 syntrophin) is an intracellular scaffold protein that promotes cancer cell proliferation, stemness, epithelial-mesenchymal transition, and metastasis by activating the Wnt/β-catenin and Hippo-YAP signaling pathways (PMID:34277816, PMID:37592763). SNTB1 forms a complex with SRC kinase and functions upstream of YAP1, whose overexpression partially rescues the proliferative and invasive defects caused by SNTB1 depletion, establishing an SNTB1–YAP1–β-catenin/MYC signaling axis in colorectal cancer (PMID:39548564, PMID:37592763). SNTB1 protein levels are post-transcriptionally upregulated through ZC3H13-mediated m6A modification of its mRNA read by YTHDF1, a mechanism that drives EMT and gastric cancer progression (PMID:40774945).

Mechanistic history

Synthesis pass · year-by-year structured walk · 4 steps
  1. 2021 Medium

Establishing that SNTB1 is not merely a structural adaptor but an active signaling scaffold: its silencing suppressed β-catenin signaling and cancer stemness, placing it functionally within the Wnt pathway in colorectal cancer.

Evidence siRNA knockdown with luciferase reporter, side-population assay, and xenograft tumorigenesis in colorectal cancer cells

    PMID:34277816

    Open questions at the time
    • Direct binding partner mediating Wnt pathway activation was not identified
    • Mechanism by which SNTB1 scaffolding activates β-catenin was not resolved
    • Findings from a single laboratory and cancer type
  2. 2023 Medium

Genetic epistasis experiments resolved the signaling hierarchy: SNTB1 acts upstream of YAP1, and YAP1 overexpression rescues SNTB1-knockdown phenotypes, linking Hippo-YAP and Wnt/β-catenin/MYC into a single SNTB1-dependent axis.

Evidence siRNA knockdown plus YAP1 rescue, colony formation, migration/invasion assays, xenograft and metastasis models in colorectal cancer cells

    PMID:37592763

    Open questions at the time
    • Molecular mechanism by which SNTB1 regulates YAP1 expression or stability was not defined
    • Whether the SNTB1–YAP1 relationship is direct or mediated through intermediate kinases was unknown
  3. 2024 Medium

Identification of SRC kinase as a direct physical partner of SNTB1 provided a candidate effector linking the scaffold to Hippo-YAP pathway activation and metastatic phenotypes.

Evidence Reciprocal co-immunoprecipitation, overexpression with in vitro proliferation/migration assays, and caudal-vein injection metastasis model in nude mice

    PMID:39548564

    Open questions at the time
    • Whether SRC kinase activity is required for SNTB1-dependent YAP activation was not tested with kinase inhibitors or catalytic-dead mutants
    • Domain mapping of the SNTB1–SRC interaction was not performed
    • Functional contribution of SNTB1–SRC complex in non-cancer contexts is unknown
  4. 2025 Medium

Upstream regulation of SNTB1 was resolved: ZC3H13-deposited m6A marks on SNTB1 mRNA are read by YTHDF1 to enhance SNTB1 protein levels, connecting epitranscriptomic regulation to EMT and gastric cancer progression.

Evidence MeRIP-seq, RNA-seq, immunoprecipitation of ZC3H13–YTHDF1–SNTB1 axis, siRNA knockdown, and in vivo functional assays in gastric cancer

    PMID:40774945

    Open questions at the time
    • Whether translational efficiency or mRNA stability is the primary mechanism of YTHDF1-mediated SNTB1 upregulation was not distinguished
    • Generalizability of the m6A regulatory axis beyond gastric cancer is untested

Open questions

Synthesis pass · forward-looking unresolved questions
  • The biochemical mechanism connecting the SNTB1–SRC complex to YAP1 activation — including whether SRC-mediated phosphorylation of Hippo pathway components is involved — remains unresolved.
  • No kinase-substrate relationship between SRC and Hippo pathway kinases (LATS1/2, MST1/2) has been tested in the context of SNTB1 scaffolding
  • Physiological, non-oncogenic functions of SNTB1 signaling through this axis are unstudied
  • Structural basis of SNTB1 scaffolding — which PDZ, PH, or syntrophin-unique domains mediate SRC and downstream interactions — is undefined
