PDLIM7 (Enigma) is a multifunctional PDZ-LIM domain scaffold protein that couples cytoskeletal organization, receptor tyrosine kinase signaling, and transcription factor trafficking to regulate cell differentiation, cardiac morphogenesis, and inflammatory responses. It anchors Ret/ptc2 to the cell periphery to enable mitogenic signaling (PMID:9528800), organizes the actin cytoskeleton through interactions with PKC and the adaptor APS (PMID:8940095, PMID:15946664), and controls nuclear–cytoplasmic shuttling of Tbx5 and YAP1 to regulate cardiac valve specification and prostate cancer growth, respectively (PMID:19895804, PMID:36823643). PDLIM7 also functions as an E3 ubiquitin ligase that polyubiquitinates the NF-κB subunit p65 and cooperates with PDLIM2 and p62/SQSTM1 to deliver it for proteasomal degradation, suppressing inflammatory signaling (PMID:32849529). In vivo, loss of Pdlim7 causes cardiac valve defects, thrombosis, and loss of vascular smooth muscle cell differentiation, establishing its requirement for cardiovascular homeostasis and hemostasis (PMID:24278323, PMID:27470516).
