Dermcidin (DCD) encodes a secreted precursor that is proteolytically processed to the antimicrobial peptide DCD-1L, which acts at membrane and biofilm interfaces in host defense (PMID:29066724, PMID:35026999). NMR analysis in membrane-mimetic micelles shows DCD-1L folds into an L-shaped molecule of three α-helices joined by flexible turns, in which charged residues of the N- and C-terminal regions carry distinct roles: the acidic C-terminal region suppresses binding to POPG membranes at basic but not acidic pH, and charged residues govern Zn2+ coordination and peptide aggregation (PMID:29066724). Functionally, DCD-1L inhibits bacterial adhesion and biofilm formation, downregulates biofilm-associated genes, and accelerates re-epithelialization in a burn-wound infection model without cytotoxic or hemolytic activity at effective concentrations (PMID:35026999). Independently, the full-length protein (designated DSEP) confers a survival advantage when overexpressed in neural cells, including resistance to oxidative stress and immune-mediated killing (PMID:12429208). Beyond these findings, the protease(s) generating DCD-1L and the molecular receptor/target of its survival activity have not been characterized in the available corpus.
