The extracellular domain of DSC1 presents conformation-dependent epitopes that serve as the autoantigen in the subcorneal pustular dermatosis (SPD) type of IgA pemphigus: pathogenic patient IgA autoantibodies bind the surface of cells expressing DSC1 but not DSC2 or DSC3, establishing isoform-specific autoimmune targeting (PMID:9242496). DSC1 expression is controlled post-transcriptionally by METTL3-mediated m6A modification, which stabilizes DSC1 mRNA, and DSC1 contributes to epithelial barrier function and limits LPS-induced apoptosis and inflammation (PMID:41453286). Beyond these findings, the structural basis of DSC1 adhesion and its detailed signaling partners have not been characterized in the available corpus.
