CAPZB encodes the beta subunit of the heterodimeric F-actin capping protein (CapZ), which binds and caps actin filament barbed ends through a C-terminal actin-binding domain containing two critical conserved leucine residues, thereby controlling filament elongation, cortical tension, and actomyosin contractility (PMID:7665558, PMID:28530659). As a stable component of the endosomal WASH complex, the CAPZA–CAPZB heterodimer facilitates Arp2/3-dependent actin nucleation on endosomes and regulates MVB trafficking for exosome secretion via ALIX-mediated recruitment (PMID:19922875, PMID:40185104). CAPZB concentrates at stereocilia tips in hair cells where mechanotransduction channel activity specifies its localization to govern stereocilia dimensions and auditory/vestibular function, and in hepatocytes it restrains ROCK–myosin-driven contractility to limit YAP activation and maintain liver size and metabolic zonation (PMID:28899994, PMID:31902726, PMID:30878582).
