ALYREF competes with the exosome cofactor hMTR4 for ARS2 binding at the cap-binding complex, thereby acting as a checkpoint that routes transcripts toward export rather than nuclear degradation (PMID:28801509). As an m5C reader, ALYREF recognizes NSUN2-deposited m5C marks to promote nuclear export and cytoplasmic stabilization of specific mRNAs including PKM2, EGFR, and JunD (PMID:28418038, PMID:33991457, PMID:38164181, PMID:38402198).
