TRIM63 (MuRF-1) is a striated muscle-specific RING zinc-finger E3 ubiquitin ligase that controls myofibrillar protein homeostasis by targeting sarcomeric proteins for degradation (PMID:11283016, PMID:15967462). It docks at the sarcomeric M-line region through a C-terminal helical domain that binds the titin A168-A170 modules adjacent to the titin kinase, with a loop protrusion in titin mediating recognition (PMID:17215480). TRIM63 binds and ubiquitinates a defined set of myofibrillar substrates—including troponin I, troponin T, myosin light chain 2, myosin heavy chain, and cardiac myosin binding protein C—routing them to proteasomal degradation; loss of TRIM63 reduces ubiquitination of MLC-2 and TnI in vivo (PMID:15967462, PMID:22821932). In cardiomyocytes, TNF-α induces TRIM63 via p42/44 MAPK to drive troponin I ubiquitination and degradation, directly impairing contractility (PMID:17145048). Beyond the sarcomere, TRIM63 mediates fasting- and denervation-induced selective autophagy of endocytosed nicotinic acetylcholine receptors at the neuromuscular junction, co-localizing with SQSTM1, SH3GLB1, and MAP1LC3A on endo/lysosomal vesicles (PMID:24220501). Its expression is transcriptionally driven by FoxO3 in cooperation with SMAD3 and by TAp63/p53, and post-transcriptionally repressed by miR-19a/b (PMID:24920680, PMID:24117217, PMID:26919175). Biallelic loss-of-function variants in TRIM63 cause autosomal-recessive hypertrophic cardiomyopathy, with mutant protein mislocalizing to Z disks, failing to auto-ubiquitinate, and failing to degrade substrates including calcineurin and mTOR, thereby activating mTOR-S6K and calcineurin hypertrophic signaling (PMID:22821932, PMID:32451364).
