Its catalytic activity requires SUMF1-mediated conversion of the active-site cysteine to Cα-formylglycine, Ca²⁺, and N-linked glycosylation, while a unique positively charged hydrophilic domain mediates high-affinity, catch-bond-dependent engagement with HS substrate (PMID:41025518, PMID:25863062, PMID:17855356, PMID:19666466).
