β-Synuclein (SNCB) is a brain-enriched presynaptic protein that contributes to dopamine homeostasis and motor behavior, acting redundantly with other synuclein family members (PMID:22128150). Loss of endogenous β-synuclein together with γ1-synuclein in zebrafish produces hypokinesia, reduced dopamine levels, and delayed dopaminergic neuron differentiation, with phenotypes rescued by human α-synuclein, establishing a phylogenetically conserved synuclein function in dopaminergic systems (PMID:22128150). At the molecular level, SNCB selectively inhibits phospholipase D2 (PLD2) and is a substrate of Ca²⁺/calmodulin-dependent protein kinase II, co-localizing with α-synuclein at presynaptic nerve terminals (PMID:9806846, PMID:11474193). When applied at elevated or extracellular concentrations to non-neuronal cells, recombinant β-synuclein engages a PLD2/p53/Mdm2 axis: in brain microvascular endothelial cells it induces dose-dependent, p53-mediated apoptosis with PLD2 activation, increased p19(Arf), and Mdm2 translocation (PMID:29808713), while in retinal pigment epithelial cells it drives PLD2-dependent p53 pathway activation alongside oxidative-stress markers HMOX1 and NOX4 (PMID:31128101). Beyond these findings, the endogenous presynaptic mechanism of SNCB has not been further characterized in the available corpus.
