SECTM1 (K12) is a type I transmembrane glycoprotein that localizes to the Golgi apparatus and also generates a soluble secreted isoform from its N-terminal domain (PMID:9480746). It functions as a costimulatory ligand for the lymphocyte receptor CD7, directly binding CD7 on T and NK cells to promote T cell proliferation, IFN-γ and IL-2 production (synergizing with CD28 signaling), and NK cell activation (PMID:10652336, PMID:22184754). SECTM1 expression is induced by IFN-γ through a STAT1-dependent pathway in monocytes and dendritic cells, establishing a positive feedback loop linking innate cytokine signals to adaptive T cell costimulation (PMID:22184754).
