SCGN

SCGN (secretagogin) is a calcium-sensor protein that facilitates regulated exocytosis in neuroendocrine cells by interacting with the SNARE complex; a missense mutation (p.Arg77His) impairs SNAP25 localization and hormone release from enteroendocrine cells (PMID:31663849). SCGN deficiency disrupts oxytocin signaling and triggers aberrant inflammatory activation, as demonstrated by genetic epistasis and pharmacological rescue with oxytocin and anti-inflammatory agents in zebrafish and mouse knockout models, with loss-of-function mutations identified in autism spectrum disorder probands (PMID:36588101).

   Establishing that SCGN is not merely a passive calcium buffer but an active participant in vesicle fusion resolved how neuroendocrine exocytosis depends on this protein: SCGN interacts with the SNARE complex, and the patient-derived p.Arg77His mutation mislocalizes SNAP25 and impairs hormone secretion.
