PLOD1 (lysyl hydroxylase 1) is an ER-resident α2 homodimeric enzyme that hydroxylates lysine residues in collagen and non-collagen substrates, thereby governing collagen crosslinking, extracellular matrix integrity, and additional signaling and structural functions in diverse cell types. The 709-amino-acid enzyme catalyzes the Fe²⁺- and 2-oxoglutarate-dependent hydroxylation of peptidyl lysine (EC 1.14.11.4) and also processes UDP-glycan donors via a distinct N-terminal glycosyltransferase domain (PMID:1577494, PMID:34400365). Loss-of-function mutations—frequently arising from Alu-mediated recombination at introns 9 and 16—cause Ehlers–Danlos syndrome type VI (kyphoscoliotic type), and PLOD1 deficiency in mice promotes aortic aneurysm through thrombospondin-1 upregulation and MMP activation (PMID:7713497, PMID:10874315, PMID:34646388). Beyond collagen, PLOD1 hydroxylates EBNA1 at K460/K461 to regulate its stability and oriP-dependent DNA replication, and binds Septin2 through its hydroxylase domain to promote actin polymerization and cancer cell confined migration (PMID:37262099, PMID:36721170).
