OSTC (also called DC2) is a subunit of the STT3A oligosaccharyltransferase (OST) complex in the endoplasmic reticulum that, together with KCP2, physically bridges the STT3A catalytic complex to the Sec61 protein translocation channel, thereby enabling efficient co-translational N-linked glycosylation; its conserved C-terminal motif mediates assembly into the STT3A complex, while its lumenal loop and N-terminal cytoplasmic segment are required for functional interaction with Sec61 (PMID:28860277). OSTC and KCP2 also interact with the γ-secretase complex and modulate amyloid precursor protein (APP) processing: their depletion causes accumulation of APP C-terminal fragments, reduces the active presenilin-1 fragment, and blocks Aβ production, whereas their overexpression stimulates γ-secretase activity and Aβ generation (PMID:21768116). OSTC expression is upregulated during the mammalian heat shock response in multiple cell types and tissues (PMID:24751383).
