ETFA encodes a mitochondrial protein that directly participates in cellular energy (ATP) production, a role demonstrated when a small-molecule PROTAC that binds and thermally stabilizes ETFA enhanced ETFA-mediated ATP generation in NSCLC cells (PMID:40815992). ETFA expression is regulated at the promoter level by multiple inputs, including EZH2-mediated promoter methylation in colorectal cancer cells (PMID:34405537) and post-transcriptional targeting of ETFA mRNA by miR-320b in endothelial cells (PMID:38556340). A homozygous ETFA promoter variant (c.-85G>A) reduces promoter activity and ETFA protein, establishing that intact ETFA expression is required to prevent multiple acyl-CoA dehydrogenase deficiency (MADD) (PMID:36579410).
