In cancer contexts, CHRDL1 suppresses proliferation, migration, invasion, and angiogenesis by inhibiting BMP receptor II–mediated Akt/Erk/β-catenin activation, TGF-β/VEGF signaling, and MAPK/MED29-driven epithelial–mesenchymal transition, while its expression is silenced by promoter hypermethylation (PMID:28423564, PMID:38415870, PMID:39462350).
