BOC binds all three mammalian Hedgehog ligands (Shh, Ihh, Dhh) directly through a specific fibronectin type III repeat, an interaction resolved at atomic resolution and shown to be essential for activity (PMID:16647304, PMID:20519495); a missense mutation in this FN3 domain abolishes BOC's signaling function in vivo (PMID:34057474). BOC forms distinct receptor complexes with Patched: a Ptch1/BOC complex signals through Gli induction, distinguishing it from the Ptch2/GAS1 complex that engages a parallel Creb/Src branch, so that the spatiotemporal balance of BOC versus GAS1 expression tunes signaling output (PMID:32332736).
